SS-31, also known as Elamipretide or MTP-131, is a synthetic aromatic-cationic tetrapeptide with the sequence D-Arg-dimethylTyr-Lys-Phe-NH₂. CAS 736992-21-5, molecular weight approximately 638.78 g/mol. The compound belongs to the Szeto-Schiller (SS) family of mitochondria-targeting peptides developed by Hazel Szeto at Cornell University.
SS-31 is designed to selectively partition into the inner mitochondrial membrane (IMM) through electrostatic and hydrophobic interactions, where it binds to cardiolipin — a phospholipid unique to and critical for the structural integrity of the IMM. Research applications include studies of mitochondrial electron transport chain function, reactive oxygen species (ROS) production, cardiolipin peroxidation, mitochondrial membrane potential dynamics, and models of mitochondrial dysfunction associated with aging and cellular stress. SS-31 is one of the most widely studied compounds in the field of mitochondria-targeted therapeutics research.
