5-Amino-1-methylquinolinium (5-Amino-1MQ) is a synthetic small molecule inhibitor of nicotinamide N-methyltransferase (NNMT), a cytosolic enzyme involved in the methylation of nicotinamide (vitamin B3) to form 1-methylnicotinamide. Molecular formula C₁₀H₁₁N₂⁺ (as cation), molecular weight approximately 159.21 g/mol as the free base. Supplied as the salt form. Note: 5-Amino-1MQ is a small molecule, not a peptide.
NNMT has emerged as a significant target in metabolic research due to its role in the regulation of the methionine cycle and cellular SAM (S-adenosylmethionine) availability. Elevated NNMT activity in adipose tissue has been associated with altered NAD⁺ metabolism, epigenetic regulation via histone methylation, and adipocyte differentiation in cell culture models. 5-Amino-1MQ is used as a selective NNMT inhibitor in research studying these connections, including adipogenesis models, metabolic reprogramming in obesity research, NAD⁺ precursor pathway biology, and the relationship between one-carbon metabolism and epigenetic gene regulation.
